Novel chromone-2-carboxylic acid derivatives, 4,10-dioxo-2-phenyl-4H,10H-benzodipyran-8-carboxylic acids (VIIa-c), have been synthesised by the condensation of appropriate 7-hydroxyflavones with dimethyl acetylenedicarboxylate (DMAD) and cyclisation of the resulting hydrolysed adduct mixtures (Va-c) with conc.sulphuric acid.The structures of these acids have been assigned on the basis of analytical and spectral data, and confirmed by their unambiguous synthesis.Among these compouds, 4,10-dioxo-3-methyl-2-phenyl-4H,10H-benzodipyran-8-carboxylic acid (VIIb) possess high antiallergic activity equivalent to that of disodium cromoglycate (DSCG) when tested by passive peritonial anaphylaxis method in rats.
