DOCUMENT TYPE: Journal CA Section: 1 (Pharmacodynamics) The effects of mafenide acetate (I) [13009-99-9] on the renal function and the electrolyte metab. were studied in pentobarbital-anesthetized dogs by i.v. injection of doses of 20,100, and 500 mg/kg. I caused a dose-dependent lowering of the systemic blood pressure and transient increase in the renal blood flow (RBF). I increased the glomerular filtration rate (GFR) at the lowest dose studied, but decreased it at higher doses. No conspicuous changes were obsd. in the filtration fraction at all doses studied. The urine flow and the Na, K and Cl-excretion in urine were increased. The increased urinary flow and electrolyte excretion were assocd. with a slight decrease in the percent reabsorption of the filtered Na and Cl. I increased the pH of urine and decreased the arterial blood pH and HCO3 level. The increase in the urinary flow and electrolyte excretion obsd. may be the result of the inhibition of the electrolyte reabsorption in renal tubules. The systemic acid-base balance was not influenced even after i.v. injection of large doses of I.
