DOCUMENT TYPE: Journal CA Section: 63 (Pharmaceuticals) Each dog was inoculated i.v. with 1.0 ′ 108 amastigotes of a Khartoum strain of Leishmania donovani/kg of body wt. The antileishmanial agents [liposome-encapsulated meglumine antimonate (I)(LEMA) [133-51-7] or unencapsulated I] were given once daily, i.v., for 1, 4, or 10 consecutive days beginning the 12th day after inoculation. A single injection of LEMA (0.61 mg Sb/kg body wt.) resulted in 89% suppression and 4 consecutive daily injections of LEMA (1.94 mg Sb/kg/day) resulted in 95.8% suppression of splenic parasites. The dose of LEMA that would give 50% suppression (SD50) was approx. 0.029 mg Sb/kg. The SD50 for unencapsulated drug was approx. 24 mg Sb/kg. The liposome-encapsulated drug was estd. to be >700-fold as effective as the unencapsulated drug. Liposomes can markedly reduce the drug dosage required for treatment of visceral leishmaniasis in dogs.

DOCUMENT TYPE: Journal CA Section: 1 (Pharmacology) Owl monkeys infested with Leishmania donovai were given meglumine antimoniate (I) [133-51-7] (6.25-500 mg Sb/kg/day, i.m.) for 10 consecutive days beginning 10 days post inoculation. I was toxic to the owl monkey at >125 mg/kg/day and I was effective therapeutically against L. donovani at dosages of 15.625-125 mg/kg/day.
