DOCUMENT TYPE: Journal CA Section: 1 (Pharmacodynamics) The opposing actions of phosphatidylserine (PS) and cromolyn (I) [16110-51-3] on histamine [51-45-6] release were studied in rat peritoneal mast cells in vitro. Histamine-releasing drugs could be sepd. into 2 groups on the basis of whether they were potentiated by PS and inhibited by .mu.M cromolyn (dextran [9004-54-0], antigen (bovine serum albumin), concanavalin A [11028-71-0]) or neither potentiated by PS nor inhibited by .mu.M cromolyn (compd. 48/80, polymyxin B [1404-26-8], phospholipase C [9001-86-9], and ATP [56-65-5]). Compd. 48/80 and polymyxin chem. combined with PS, but this could be circumvented by preincubating the mast cells in PS for 1 h, and then washing them free of PS in soln. prior to addn. of the drugs. These data define at least 2 pathways for induction of histamine release: a PS-potentiated pathway inhibited by .mu.M cromolyn, and a PS-independent pathway unaffected by .mu.M cromolyn. Dose-response curves of the .mu.M cromolyn inhibition of PS-potentiated release revealed a parallel shift, suggesting that cromolyn may compete with PS.

These results may have relevance to the mechanism of action of the antiallergic cromolyns on mast cells. .
