Phosphodiesterase inhibitors, cAMP analogs and b-adrenoceptor agonists variously blocked release of the amine from all 3 cell types. The mouse cells were rather less sensitive to the former agents than those of the rat and hamster. Cromoglycate [15826-37-6] inhibited histamine secretion from rat peritoneal mast cells, was less active against the hamster cells and completely ineffective against those of the mouse. Other chromones showed varied patterns of differential reactivity but phloretin [60-82-2] and the flavonoid quercetin [117-39-5] were essentially equiactive against all 3 cell types. The tetrazoles AH 9679 [81859-17-8] and doxantrazole [51762-95-9] prevented histamine release in each case but were less active against the mouse cells. These results further emphasize the functional heterogeneity of mast cells from different sources. .

DOCUMENT TYPE: Journal CA Section: 1 (Pharmacology) The effect of N-(3,4-dimethoxycinnamoyl)anthranilic acid (N-5') [53902-12-8], an orally applicable anti-allergic drug, on the histamine [51-45-6] release induced by calcium ionophores A23187 [52665-69-7] and X537A [25999-31-9] and by ATP [56-65-5] from rat peritoneal exudate cells (PEC) was studied. X537A (0.1-33.3 mg/mL) induced histamine release in a concn.-related manner, and 2.0 mg/mL of X537A induced release to the same extent as allergic histamine release. Histamine release induced by 2.0 mg/mL of X537A increased with longer incubation time, reaching a peak of about 100% at 120 min. N-5' had no effect on histamine release induced by X537A at the concns. used (1-1000 mM), but disodium cromoglycate (DSCG) [15826-37-6] exhibited significant inhibition at 1-100 mM. A23187 (0.05-0.5 mg/mL) induced histamine release in a concn.-related manner, and it seemed the 0.033 mg/mL at A23187 induces the same degree of histamine release as in allergic release. A23187 induced rapid histamine release which attained max. 1 min after the addn.In this experiment, several chemicals are used like 51-45-6 and 53902-12-8 N-5' exhibited a significant concn.-dependent inhibitory effect on histamine release induced by A23187, and DSCG also exhibited significant inhibition (10 and 1000 mM). N-5' significantly inhibited histamine release induced by 100 mM ATP. These results indicate that N-5' and DSCG effect the histamine release induced by ionophore A23187 and X537A by different manners, and they suggest the possibility that N-5' inhibits some Ca2+-dependent processes in histamine release, including the influx of Ca2+ into cells, which is a trigger of the A23187 and ATP effects. .
