DOCUMENT TYPE: Journal CA Section: 4 (Toxicology) The first step in the metab. of a polycyclic hydrocarbon mol. is the satn. of a double bond, the formation of an epoxide, then of a dihydrodiol. Once sepn. has been achieved by chromatog., the absorption spectra of these dihydrodiols can be established and directly compared with theor. predictions which are reasonably easy to elaborate in the case of planar mols. The initial site of enzyme attack can then be identified without using model mols. Absorption and fluorescence spectra of pyrene [129-00-0], benzo[e]pyrene [192-97-2], benzo[a]pyrene [50-32-8], dibenzo[a,e]pyrene [192-65-4] and some of their dihydrodiol metabolites are given as examples.

DOCUMENT TYPE: Journal CA Section: 4 (Toxicology) Examn. of the values obtained for polarog. half-wave potentials for 22 carcinogenic and noncarcinogenic polycyclic compds. indicated that carcinogens such as 3,4-benzopyrene (I) [50-32-8], 1,2,4,5-dibenzopyrene [192-65-4], 3,4,8,9-dibenzopyrene [189-64-0], and 3,4,9,10-dibenzopyrene [189-55-9] were reduced with more pos. values of 1st and 2nd wave potentials than were their corresponding inactive isomers, 1,2-benzopyrene [192-97-2] and 1,2,6,7-dibenzopyrene [192-51-8]. The possible relation between redn. mechanisms and carcinogenic activity are discussed.
