Vitamin A metabolism: analysis of steady-state neutral metabolites in rat tissues
Vitamin A metabolism: analysis of steady-state neutral metabolites in rat tissues. Williams, John B.; Pramanik, Bikash C.; Napoli, Joseph L. (Health Sci. Cent., Univ. Texas, Dallas, TX 75235, USA). J. Lipid Res., 25(6), 638-45 (English) 1984. CODEN: JLPRAW. ISSN: 0022-2275. DOCUMENT TYPE: Journal CA Section: 18 (Animal Nutrition) Section cross-reference(s): 9, 13 High-performance liq. chromatog. systems were developed to rapidly sep. retinol from its esters, analyze the total spectrum of neutral vitamin A [68-26-8] compds., and purify retinyl esters to homogeneity. Chem. ionization mass spectrometric techniques were used to identify vitamin A compds.; these techniques are also applicable to quantification of tissue vitamin A compds. These methods provide rapid and sensitive techniques for sepn. and quantification of neutral retinol metabolites. Their utility was demonstrated by anal. of vitamin A metabolites in rat tissues under steady-state conditions. Tissue specificity was noted for the concns. of retinol and its long-chain fatty acid esters, the ratio of retinol to retinyl esters, and the fatty acid compn. of retinyl esters. Quant. minor amts. of several neutral polar retinol metabolites were detected, but neither 13-cis-retinol [2052-63-3] nor 4-hydroxyretinol [15353-44-3] was obsd. in vivo as metabolites of retinol in kidney.

Studies of the improvement of drug analysis: HPLC procedure for the simultaneous determination of retinol, a-tocopherol, their esters, b-carotenes, and other carotenoids. Cavina, G.; Gallinella, B.; Pecora, P.; Suraci, C.; Porra, R. (Inst. Super. Sanita, Rome 299, Italy). Boll. Chim. Farm., 122(11), 531-48 (Italian) 1983. CODEN: BCFAAI. ISSN: 0006-6648. DOCUMENT TYPE: Journal CA Section: 18 (Animal Nutrition) Section cross-reference(s): 1, 9 The sepn. and detn. of trans-retinol [68-26-8], dl-a-tocopherol [10191-41-0], and b-carotene [7235-40-7] and their esters as well as 13-cis-retinol [2052-63-3] from human blood serum was accomplished by a reversed-phase HPLC method (LiChrosorb RP column; eluant MeOH, MeCN, or MeCN-H2O ). The method is reliable and precise with coeff. of variation 2.0-4.8%. The procedure allows addnl. detn. of serum carotenoids by repeating sample ext. injection, and monitoring the effluent at a suitable wavelength.
