DOCUMENT TYPE: Journal CA Section: 1 (Pharmacodynamics) Buflomedil (I) [35543-24-9], a new peripheral vasodilator, was detd. in the serum of persons receiving 100 mg i.v. or 450 mg orally by extn. of the deproteinized serum at pH 10.2 with Et2O and gas chromatog. of the ext., using a thermoionic detector. Various pharmacokinetic parameters of I was calcd. and indicated a tissue distribution, with a vol. of the peripheral compartment twice that of the central compartment. The biol. half-lives of I were similar after i.v. and oral administration: 1.91 and 2.10 h, resp. After i.v. injection the total distribution vol. of I was 40 L, the total clearance 292 mL/min, and the half-life of elimination 0.6 h. After an oral dose of 450 mg the half-life of absorption was 0.17 h and the max. blood concn. 3.8 mg/L. The abs. bioavailability of the oral form with respect to the i.v. route was 30%, explainable by a 1st-pass effect.
