Thus, Corynebacterium N-774 was shake-cultured at 30° for 3 days in a pH 7.2 medium contg. glucose 10, peptone 5, yeast ext. 3, and malt ext. 3 g/L. The broth was centrifuged and the cells washed with phosphate buffer. The wet cells (0.5 g, ~80% H2O) were added to 100 g 0.05M phosphate buffer (pH 8.5) contg. 2% b-hydroxypropionitrile (I) [109-78-4] and treated at 30° for 1 h with stirring. The resulting mixt. contained 2.5% b-hydroxypropionamide (II) [2651-43-6] and no b-hydroxypropionic acid [503-66-2]. The reaction mixt. (1 L) was centrifuged and the supernatant concd. and the residue extd. with MeCN to give a 75.8% yield of II. Similarly, b-hydroxypropionic acid was prepd. from I by incubation at 30° for 24 h. Also prepd. were b-chloropropionic acid [107-94-8] and its amide [5875-24-1], b-mercaptopropionic acid [107-96-0] and its amide [763-35-9], and N,N'-dimethyl-b-aminopropionamide [50836-82-3]. .

The most abundant hydroxy acid in black kraft liquor was 2-methylol-2,4,5-trihydroxypentanoic acid [66702-03-2]. .
