Acetylcholine [51-84-3] induced a dose-dependent vasodilation. dl-Norepinephrine [138-65-8] and dl-epinephrine [329-65-7] produced a vasoconstriction followed by a strong vasodilation in large coronary arteries and only a weak vasodilation in small coronary arteries. Phenylephrine [59-42-7] (a selective a1-agonist) induced a strong vasoconstriction in both arteries. The threshold dose and 50% ED value for phenylephrine in small coronary arteries were much larger than those in large coronary arteries, although the vasoconstrictions by KCl and PGF2a [551-11-1] were not different between in large and small coronary arteries. Clonidine [4205-90-7] and xylazine [7361-61-7] (selective a2-agonists) produced a slight vasoconstriction but not dose dependently and a vasodilation with extremely large doses. The 50% ED value of vasodilation for salbutamol [18559-94-9] (a selective b2-agonist) was ~80-fold greater than that for l-isoproterenol [51-31-0] (nonselective b-agonist) in large coronary arteries, but was ~20-fold in small coronary arteries. The maximal dilator response to salbutamol was about the same as that to isoproterenol in small coronary arteries, whereas it was much smaller than that to isoproterenol in large coronary arteries. Evidently, adrenoceptors are heterogeneous according to the distance from the coronary office in canine epicardial coronary arteries. .

DOCUMENT TYPE: Journal CA Section: 1 (Pharmacology) The ability of tolazoline [59-98-3] and yohimbine [146-48-5] to antagonize xylazine [7361-61-7]-induced central nervous system depression, bradycardia, and tachypnea in 9 ewes and 5 rams was studied.In this article, certain chemicals are used. Some of their cas registry numbers are 59-98-3 and 146-48-5 Once a week of 3 wk, each sheep received one i.v. treatment of 0.4 mg xylazine/kg, 0.4 mg xylazine/kg followed in 10 min by 2 mg tolazoline/kg, or 0.4 mg xylazine/kg followed in 10 min by 0.2 mg yohimbine/kg. The order of the 3 treatments in each sheep was randomized. Xylazine alone caused recombency for 41.0 min. Tolazoline and yohimbine shortened the xylazine-induced recumbency to 12.1 min and 18.1 min, resp. Sheep given xylazine alone had head droop for 34.0 min after rising. Head drooping of sheep given tolazoline or yohimbine was reduced to 10.1 min and 14.2 min, resp. Both tolazoline and yohimbine reversed the bradycardia and tachypnea that followed xylazine administration. No statistical differences in the rate and magnitude of the reversal were obsd. between the 2 drugs. .
