Caspase activator
A caspase activator is a substance or compound that stimulates the activity of caspases, which are key enzymes involved in programmed cell death (apoptosis). Caspases are cysteine proteases that exist in cells as inactive proenzymes. They play essential roles in executing apoptotic pathways by cleaving specific cellular proteins, leading to cell dismantling and death.Caspase activation is tightly regulated and occurs through various mechanisms depending on the apoptotic stimulus. Caspase activators can act upstream of caspases by inducing their cleavage from their proenzymatic form to their active form. They can also promote the assembly of multiprotein complexes, such as the apoptosome in the intrinsic apoptotic pathway or the death-inducing signaling complex (DISC) in the extrinsic pathway, which in turn activate caspases. Additionally, some activators modulate the mitochondrial membrane permeabilization, releasing apoptogenic factors that trigger caspase activation.
