Hedgehog / Smoothened inhibitor

Hedgehog/Smoothened inhibitors are compounds that target and block the activity of the Smoothened (SMO) receptor, a component of the Hedgehog signaling pathway critical for embryonic development and tissue regeneration in adults. This pathway plays a crucial role in regulating cell growth, differentiation, and patterning during embryogenesis and maintains tissue homeostasis in adulthood. Dysregulation of Hedgehog signaling, often through mutations in SMO or other pathway components, is implicated in various cancers, including basal cell carcinoma and medulloblastoma. Hedgehog/Smoothened inhibitors act by preventing SMO activation and downstream pathway activation, thereby inhibiting tumor cell proliferation and promoting apoptosis.
