5-Acyl-2-acylamino-1H-benzimidazoles (3a-w) have been synthesized and evaluated for their macrofilaricidal and chemosterilizing activities in experimental animals.Compounds 3a,b,q exhibit high order of adulticidal activity against L. carinii (cotton rats) and A. viteae (Mastomys) infections while compounds 3b,o q show marked chemosterilizing effect on adult female filariids.

The synthesis of a series of 2-substitutedamino-5(6)-benzoylbenzimidazoles (6-22) has been carried out as the structural analogs of 2-amino-5(6)-benzoyl/α-hydroxy-α-phenylmethylbenzimidazoles (3, 4), the two metabolites of mebendazole.The compounds 6-22 have been tested for their anthelmintic activity against Ancylostoma ceylanicum, Nippostrongylus brasiliensis and Hymenolepis nana in rodents.Compounds 9, 11, 12, 13 and 14 exhibit 100percent elimination of A. ceylanicum worms from the animals at a single oral dose of 50-250 mg/kg.Some of the compounds also show activity against H. nana and N. brasiliensis.
