The invention discloses a method of preparing 3-substituted-2-bromo-6-trifluoromethyl pyridine. The method comprises the following steps: in the presence of nitrogen, reacting 2-bromo-6-trifluoromethyl pyridine used as a raw material with lithium diisopropylamide at a low temperature to generate 3-site lithium salt, and then adding trimethyl borate, ethyl formate and carbon dioxide respectively to generate 2-bromo-6-trifluoromethyl-3-pyridineboronic acid, 2-bromo-6-trifluoromethyl-3-pyridylaldehyde and 2-bromo-6-trifluoromethyl-3-picolinic acid; and reducing 2-bromo-6-trifluoromethyl-3-pyridylaldehyde by sodium borohydride to generate 2-bromo-6-trifluoromethyl-3-pyridinemethanol. The generated target compound is high in yield and good in purity, is an important medical intermediate, and has a wide application prospect. The method disclosed by the invention is simple and convenient to operate, easily available in raw materials, good in reaction selectivity, environment-friendly, easy for enlarged production, and capable of realizing commercialized products.
