Recently, we have developed a method for catalytic regioselective synthesis of 2-substituted and 3-substituted benzofurans starting from phenols. The choice of reacting partner, olefin versus α,β-unsaturated acid, is critical to dictate the isomeric product formation. Instances are known where these olefinic partners did not complement each other and yield a similar outcome. In the current work, we have addressed this paradox with emphasis on (a) the origin of orthogonal selectivity and (b) the key requirements to expect complementary behavior. Experimental and computational studies provided important mechanistic insights. Electrostatic compatibility during migratory insertion and the positioning of the carboxylic acid moiety in catalytic steps are found to exert a paramount impact in determining the regioselectivity. The study offers a predictable single component tuning tool to control the regioselectivity.

A tandem chemoselective synthesis of 2-arylbenzofurans was accomplished from o-hydroxy-gem-(dibromovinyl)benzenes and BiAr3 reagents under palladium-catalyzed conditions. This unique and synthetically valuable strategy proceeds through three consecutive coupling reactions involving triarylbismuth reagents and provides 2-arylbenzofuran products in high yields. This paper describes the preparation of indoles, azaindoles and benzofurans in pure water by using a new heterogeneous Pd-Cu/C catalyst through a cascade Sonogashira alkynylation-cyclization sequence. Details of the optimization studies and the substrate scope are discussed. This procedure allows the preparation of heterocycles with good yields and is tolerant to a wide variety of functional groups.

The palladium catalyzed, atom-economic synthesis of various functionalized 2-arylbenzofurans was achieved through cross-coupling reaction of 2-bromobenzofurans with triarylbismuth reagents. The palladium catalytic protocol is very efficient to furnish various cross-coupled functionalized 2-arylbenzofurnas in high yields using triarylbismuth reagents with three aryl couplings as multi-coupling organometallic nucleophiles in one-pot operation. All the coupling reactions were completed in 1 h short reaction time involving three couplings from triarylbismuths under heating condition.

A solventless, microwave-enhanced Sonogashira coupling reaction of aromatic iodides with terminal alkynes on potassium fluoride doped alumina in the presence of palladium powder, cuprous iodide, and triphenylphosphine has been developed. The reaction can be utilized to prepare aryl alkynes in excellent yields. The coupling of o-iodophenol with terminal alkynes leads to the formation 2-substituted-benzo[b]furans. Whereas the coupling of o-iodoanilines with terminal alkynes generates indole products. An in situ desilylation reaction was also developed.

Reaction of phenols with 2-(isopropylthio)ethyl acetate activated by sulphuryl chloride afforded 2-phenols regioselectively, via sigmatropic rearrangement of phenoxysulphonium ylides.The ortho-alkylated phenols thus obtained have been cyclized with conc. hydrochloric acid in 2-methoxyethanol to 1-benzofurans. 2-Methyl- and 2-phenyl-1-benzofurans have been prepared similarly.
