The invention relates to the field of pharmacy, and particularly relates to a preparation method of an acetyl-coA carboxylase inhibitor. The method comprises the following steps of firstly enabling 2-(6-halo-5-methyl-2,4-dioxo-1,4-dihydrothieno[2,3-d]pyrimid-3(2H)-yl)-2-tert-butyl methacrylate (II) to react with halogenated o-methoxyacetophenone, then hydrolyzing, protecting a carboxyl group witha silicon group, reducing, halogenating, subsequently substituting a halogen atom by alcohol to obtain a key intermediate 2-(6-halo-1-(2-(2-methoxyphenyl)-2-alkoxy)-ethyl)-5-methyl-2,4-dioxo-1,4-dihydrothieno[2,3-d]pyrimid-3(2H)-yl)-2-silicon methacrylate (VIII), and finally preparing a target compound through Stille coupling and the removal of a protecting group of the silicon group. The method is a few in side reaction, low in cost, simple and convenient to operate and suitable for industrialized production.
