Over 50 tetrahydroindazoles were synthesized after 7-bromo-3,6,6-trimethyl-1-(pyridin-2-yl)-5,6,7,7a-tetrahydro-1H-indazol-4(3aH)-one (3) was identified as a hit compound in a high throughput screen for inhibition of CDK2 in complex with cyclin A. The activity of the most promising analogues was evaluated by inhibition of CDK2 enzyme complexes with various cyclins. Analogues 53 and 59 showed 3-fold better binding affinity for CDK2 and 2- to 10-fold improved inhibitory activity against CDK2/cyclin A1, E, and O compared to screening hit 3. The data from the enzyme and binding assays indicate that the binding of the analogues to a CDK2/cyclin complex is favored over binding to free CDK2. Computational analysis was used to predict a potential binding site at the CDK2/cyclin E1 interface.

The invention belongs to the field of medicines, and particularly relates to a 2-(granatane3-amino)-4-tetrahydronaphthalene indazole-substituted benzamide compound and an application thereof. The 2-(granatane3-amino)-4-tetrahydronaphthalene indazole-substituted benzamide compound has a structure shown in a formula I in the specification, wherein R1 is a hydrogen group or an alkyl group and the like; R2 is a hydrogen group, an alkyl group or a hetero-atom-substituted alkyl group and the like. The compound is obtained by substituting the second site of 4-tetrahydronaphthalene indazole-substituted benzamide with granatane-3-amino. Growth of a plurality of tumor cells can be inhibited; the tumor cells can be induced to move towards an apoptosis channel; and the 2-(granatane3-amino)-4-tetrahydronaphthalene indazole-substituted benzamide compound has potential application value in the fields such as tumor treatment and auxiliary treatment.

The μ-opioid receptor (MOR) is the major opioid receptor targeted by most analgesics in clinical use. However, the use of all known MOR agonists is associated with severe adverse effects. We reported that the 1-phenyl-3,6,6-trimethyl-1,5,6,7-tetrahydro-4H-indazol-4-ones are novel opioid receptor agonists. Subsequent structural modification resulted in the potent MOR/KOR (κ-opioid receptor) agonists 19, 20, and 21. Testing the analgesic effect of these in WT B6 mice (tail-flick test) gave ED50 values of 8.4, 10.9, and 26.6 mg/kg, respectively. The 1-phenyl-3,6,6-trimethyl-1,5,6,7- tetrahydro-4H-indazol-4-one core could be addressed in 1 or 2 synthetic steps with moderate to high percent of yield. In the adenylyl cyclase assay, compound 19 displayed a MOR/KOR agonist profile, with IC50 values of 0.73 and 0.41 μM, respectively. Current results suggest that compound 19 is a promising lead to go further development and in vitro/in vivo adverse effects studies.

Fused isoxazoles underwent reductive ring-opening in the presence of molybdenum hexacarbonyl to give the corresponding β-disubstituted compounds. 3,6,6-Trimethyl-6,7-dihydro-1,2-benzisoxazol-4(5H)-one oxime underwent reductive ring-opening in the presence of molybdenum hexacarbonyl to give 3,6,6-trimethyl-6,7-dihydro-1H-indazol-4(5H)-one. A mechanism is proposed.
