Synthesis and Properties of 2'-O-Methoxymethyl Oligonucleotides

2'-O-Methoxymethyluridine was synthesized under mild conditions using N-iodosuccinimide (NIS) activation of a 2'-methoxythiomethyl function in presence of methanol.Oligoribonucleotides containing such modified uridine residues had slightly decreased duplex stability when compared with full RNA duplexes.No substantial increase in stability towards snake venom phosphodiesterase (SVPD) was observed.
