Synthesis, Antiproliferative and Antioxidant Activity of 3-Mercapto-1,2,4-Triazole Derivatives as Combretastatin A-4 Analogues

Two series of 3-mercapto-1,2,4-triazole derivatives containing alkoxy substituents different in size and position were synthesized and their structures were characterized by FT-IR, 1H NMR, 13C NMR spectroscopy and elemental analysis. The synthesized compounds were assessed for their antiproliferative activity against colon cancer cell line (SW480). The results indicated that the size and position of the alkoxy group significantly influenced the antiproliferative activity. The highest cancer cell growth inhibition values were observed for the compounds containing 3,4,5-trimethoxyphenyl groups in their structures (57.74, 54.14 and 60.70% at 50 μM for compounds 5a, 12b and 14, respectively). The synthesized compounds were also subjected to DPPH protocol for evaluating the antioxidant activity. The results showed that all compounds had moderate to high levels of antioxidant capacity as compared to ascorbic acid as standard, the highest free radical scavenging capacity of 75% was observed for compound 4a at 50 μM.

We have developed a method for the esterification of carboxylic acids with alcohols using amphipathic, monolithic-resin bearing sulfonic acid moieties as cation exchange functions (monolith-SO3H). Monolith-SO3H efficiently catalyzed the esterification of aromatic and aliphatic carboxylic acids with various primary and secondary alcohols (1.55.0 equiv) in toluene at 6080 °C without the need to remove water generated during the reaction. The amphipathic property of monolith-SO3H facilitates dehydration due to its capacity for water absorption. This reaction was also applicable to thioesterification, wherein the corresponding thioesters were obtained in excellent yield using only 2.0 equiv of thiol in toluene, although heating at 120 °C was required. Moreover, monolith-SO3H was separable from the reaction mixtures by simple filtration and reused for at least five runs without decreasing the catalytic activity.

A mild and green electro-oxidative protocol to construct aromatic esters from methylarenes and alcohols is herein reported. Importantly, the reaction is free of metals, chemical oxidants, bases, acids, and operates at room temperature. Moreover, the design of the electrolyte was found critical for the oxidation state and structure of the coupling products, a rarely documented effect. This electro-oxidative coupling process also displays exceptional tolerance of many fragile easily oxidized functional groups such as hydroxy, aldehyde, olefin, alkyne, as well as neighboring benzylic positions. The enantiomeric enrichment of some chiral alcohols is moreover preserved during this electro-oxidative coupling reaction, making it overall a promising synthetic tool.

C(OH)–C bonds are widely distributed in naturally renewable biomass, such as carbohydrates, lignin, and their platform molecules. Selective cleavage and functionalization of C(OH)–C bonds is an attractive strategy in terms of producing value-added chemicals from biomass. However, effective transformation of alcohols into esters by activation of C(OH)–C bonds has not been achieved so far. Herein, for the first time, we report selective cleavage and esterification of C(OH)–C bonds, catalyzed by inexpensive copper salts, using environmentally benign oxygen as the oxidant, to afford methyl esters in excellent yields. A diverse range of phenylethanol derivatives that contain C(OH)–C bonds were effectively converted into methyl benzoates. Detailed analysis revealed that the high efficiency and selectivity resulted mainly from the fact that, in addition to the major esterification reaction, the side products (e.g., olefins and acids) were also transformed in situ into esters in the reaction system. C(OH)–C bonds are widely distributed in naturally renewable biomass. In the context of developing future biorefineries, selective cleavage and functionalization of C(OH)–C bonds are crucial and represent an attractive strategy in terms of producing value-added chemical compounds from biomass resources. In the current manuscript, we report, for the first time, an effective and selective method for the cleavage and esterification of C(OH)–C bonds of alcohols to produce esters, by using environmentally benign O2 as the terminal oxidant and inexpensive commercially available copper salts as catalysts. Furthermore, a detailed mechanistic study revealed that, in addition to the major esterification route, side products (e.g., olefins and acids), which are inevitably generated under oxidative and basic conditions, were also simultaneously converted into esters, thus significantly improving the final yields of target ester products. Native lignin represents the only naturally sustainable aromatic resource. Transformation of native lignin into valuable aromatics would make a great contribution to our planet. We report, for the first time, the effective transformation of alcohols into esters by esterification of C(OH)–C bonds, which offers a new way for the simultaneous degradation and functionalization of lignin. This reaction promotes new explorations for biomass valorization.
Ruthenium-catalyzed hydrogenation of CO2as a route to methyl esters for use as biofuels or fine chemicals

A novel robust diphosphine-ruthenium(ii) complex has been developed that can efficiently catalyze both the hydrogenation of CO2 to methanol and its in situ condensation with carboxylic acids to form methyl esters; a TON of up to 3260 is achievable for the CO2 to methanol step. Both aromatic and aliphatic carboxylic acids can be transformed to their corresponding methyl esters with high conversion and selectivity (17 aliphatic and 18 aromatic examples). On the basis of a series of experiments, a mechanism has been proposed to account for the various steps involved in the catalytic pathway. More importantly, this approach provides a promising route for using CO2 as a C1 source for the production of biofuels, fine chemicals and methanol.

Two homologous series based on three linking groups have been synthesized and well characterized by elemental analyses and spectroscopic techniques such as Fourier transform infrared [FT-IR] and proton magnetic resonance magnetic resonance [1H NMR] spectroscopy. The mesomorphic properties of these compounds were observed by using optical polarized microscopy (POM) and confirmed by differential scanning calorimetry (DSC) analysis. In this present investigation, we have synthesized two homologous series viz. (E)-4-(3-(4-(tetra decanoyloxy) phenyl) acryloyl) phenyl-4-n-alkoxy benzoate (Series-1) and 4-((E)-3-(4-(((E)-3-(4-n-alkoxy phenyl) acryloyl) oxy) phenyl)-3-oxo prop-1-en-1-yl) phenyl tetradecanoate (Series-2). Both of the series are differing with respect to the first linking group. All the homologous in following series displays LC properties on heating as well as cooling condition except first four homologous (C1 to C4) in series-1 and six homologous (C1 to C6) in series-2. To get more insights, the HOMO, LUMO studies are carried out which supports intramolecular charge transfer interactions in this class of mesogens.

In this present article, we wish to report on the liquid-crystalline properties of chalcone-ester based homologous series of compounds having aliphatic chain length in n-alkoxy group (n = 1 to 10, 12, 14, 16, 18). The present series consisted thirteen compounds, in which comp.C1 and C2 shows nonliquid crystalline properties, while comp.C3 to C7 display SmC phase and comp.C8 to C18 exhibits only nematic phase. Textural pattern of presently synthesized compounds are schlieren and threaded type. All this compounds were well characterized by elemental analysis, FT-IR and 1H NMR. Phase transition temperatures of present synthesised compounds were determined by optical polarising microscopy (POM), differential scanning calorimetric (DSC). Detailed XRD investigation endorses the presence of the nematic phase in higher homologues and SmC phase in lower homologues. It is shown that chalcone with ester as linking unit favors a calamitic liquid crystalline behaviour in molecules. To get more insights, the DFT based HOMO, LUMO studies are carried out which supports intramolecular charge transfer interactions in this class of mesogens. Chalconyl ester based compounds C3 to C12 shows antibacterial as well as antifungal activity compared with corresponding standard drugs.
Identification of new aryl hydrocarbon receptor (AhR) antagonists using a zebrafish model

A new series of 1,3-diketone, heterocyclic and α,β-unsaturated derivatives were synthesized and evaluated for their AhR antagonist activity using zebrafish and mammalian cells. Compounds 1b, 2c, 3b and 5b showed significant AhR antagonist activity in a transgenic zebrafish model. Among them, compound 3b, and 5b were found to have excellent AhR antagonist activity with IC50 of 3.36 nM and 8.3 nM in a luciferase reporter gene assay. In stem cell proliferation assay, compound 5b elicited marked HSC expansion.
Nitrogen Dioxide Catalyzed Aerobic Oxidative Cleavage of C(OH)–C Bonds of Secondary Alcohols to Produce Acids

Stable organic nitroxyl radicals are an important class of catalysts for oxidation reactions, but their wide applications are hindered by their steric hinderance, high cost, complex operation, and separation procedures. Herein, NO2 in DMSO is shown to effectively catalyze the aerobic oxidative cleavage of C(OH)?C bonds to form a carboxylic group, and NO2 was generated in situ by decomposition of nitrates. A diverse range of secondary alcohols were selectively converted into acids in excellent yields in this transition-metal-free system without any additives. Preliminary results also indicate its applicability to depolymerize recalcitrant macromolecular lignin. Detail studies revealed that NO2 from nitrates promoted the reaction, and NO2 served as hydrogen acceptor and radical initiator for the tandem oxidative reaction.
New chalcone based liquid crystals with allylidene amino linking unit: Synthesis and characterization

In this study, the synthesis, structural characterization and mesomorphic properties of newly thirteen calamitic shaped compounds derived from allylidene amino chalcone and 4-n-alkoxy benzoyloxy benzoic acid. Comp.H1 to H3 exhibited nonliquid crystalline nature, while comp.H4 to comp.H18 displayed enantiotropical smectic C phase. Phase transition temperatures of present synthesised compounds were determined by optical polarising microscopy (POM), differential scanning calorimetric (DSC) and powder X-ray diffraction (PXRD) techniques. Thermal stabilities of smectic to isotropic phase are 150.0 °C and temperature range of mesophase is in decreasing order from comp.H3 to comp.H18 respectively. The presences of SmC phase are the type of broken fan and needle type in present synthesized series. It is also shown that presence of chalcone amino allylidene central linking group favors a calamitic liquid crystalline behaviour in molecules with lower member to higher member aliphatic side chain in alkoxy group (-OR).
