A continuous end-to-end synthesis and purification of diphenhydramine hydrochloride featuring atom economy and waste minimization is described. Combining a 1:1 molar ratio of the two starting material streams (chlorodiphenylmethane and N,N-dimethylaminoethanol) in the absence of additional solvent at high temperature gives the target compound directly as a molten salt (ionic liquid above 168 °C) in high yield. This represents the first example of continuous active pharmaceutical ingredient (API) production in this manner. Six of the twelve principles of green chemistry as defined by the American Chemical Society are achieved, most prominently waste minimization and atom economy.

The effects of temperature and pressure on the reversible reaction between GH and aliphatic amines in acetonitrile have been studied spectrophotometrically.The formation of the free ions rather than that of ion pairs has been confirmed by the modified Benesi-Hildebrand (B-H) plot.From the plot the equilibrium constants were estimated.From the temperature dependence of the equilibrium constant (K), the heat and entropy of reactions were estimated to be: (ΔH0)av=-44.5 kJ mol-1, and (ΔS0)av=-193 J K-1 mol-1.From the pressure dependence of K the reaction volume (ΔV0)av was estimated to be -40 dm3 mol-1.When unbranched primary amines were used, a side reaction took place for which an explanation has been given.

The kinetics of hydrolysis of meclofenoxate hydrochloride (MF-HCl) in commercial tablets were studied in comparison with those of the pure solid. MF-HCl in tablets was found to be decomposed by water vapor in the same way as the pure solid. At a humidity above the critical relative humidity (CRH) of the system, the degradation ratio, x could be correlated to time by the equation: x = kt(n) where k and n are parameters. At humidities below the CRH, on the other hand, the degradation did not conform to this equation because of the fast decomposition at the initial stage. The stability of several commercial MF-HCl tablets was studied, and some excipients such as magnesium carbonate were found to reduce the stability of the drug.
