Facile synthesis of 3(2H)-furanones

Abstract: A practical method for the synthesis of 3(2H)-furanones including the bullatenone was described. Intramolecular cyclization of 4-hydroxyalkynones in the presence of KOH affords the biologically potent furanones in moderate-to-good yield at room temperature. Synthesis of 4-hydroxyalkynones from the reaction of acid chloride and terminal alkyne in the presence of copper iodide at room temperature was also reported.

An efficient approach to tetrasubstituted alkenylboronates via a cascade borylation/B-O elimination of propynols and B2pin2 was disclosed. A series of tetrasubstituted alkenylboronates were readily furnished with this strategy in good yields, with further transformations leading to tetrasubstituted alkenes and β-diketones demonstrating the synthetic potential of the alkenylboronates constructed by this strategy as versatile intermediates in organic synthesis.

PROBLEM TO BE SOLVED: To provide a perfluoroalkylphosphine compound, and a complex between a metal and the perfluoroalkylphosphine. SOLUTION: A perfluoroalkylphosphine compound is a phosphine compound represented by the general formula Rf-PR1R2. In the formula, R1 and R2 are each independently a substituted or unsubstituted hydrocarbon group; Rf is a perfluorinated hydrocarbon group. PROBLEM TO BE SOLVED: To provide methods of producing a perfluoroalkylphosphine compound improved in a phosphorus element yield and a chemical yield. SOLUTION: A method of producing a perfluoroalkylphosphine compound comprises either reacting a perfluoroalkyl iodide with, e.g., 2,4,6-trimethylbenzoyldiphenylphosphine oxide (TMDPO) or a triarylphosphine such as triphenylphosphine, which are easily available, in the presence of a radical generator such as azobis(isobutyronitrile) or light irradiation, or reacting TMDPO or the like with perfluoroalkyl iodide in the presence of diphenylphosphine, diethylphosphine, dicyclohexylphosphine or the like. A practical synthesis yielding P-perfluoroalkylated phosphines from triarylphosphines and perfluoroalkyl iodides has been developed. The photoinduced reaction involves the substitution of aryl groups on the phosphorus atom with perfluoroalkyl groups to successfully afford P-perfluoroalkylated phosphines. In addition, the P-perfluoroalkylated phosphines were found to promote the Cu-free cross-coupling reaction of acid chlorides with terminal alkynes.

A carbonylative three-component coupling reaction of aryl iodides with terminal alkynes catalyzed by PdCl2(PPh3)2 was carried out using an ionic liquid, [bmim]PF6, as the reaction medium, which resulted in good yields of α,β-acetylenic ketones. The low-viscosity ionic liquid, [bmim]NTf2, was not suitable for this reaction, since the background Sonogashira coupling reaction, a competing reaction, also proceeded.

Polymethylhydrosiloxane (PMHS) in combination with CsF facilitates the cross-coupling of alkynes or benzothiazoles with an array of vinyl, styryl, and aryl halides or nonaflates as well as acid chlorides. Experimental and spectroscopic evidence indicates that these reactions involve the in situ generation of a siloxyl intermediate. These cross-couplings proceed relatively quickly at room temperature and under amine-free conditions. To demonstrate the applicability of the method, a total synthesis of the cyctotoxic butanolide (-)-akolactone A was carried out.
