The invention discloses a progesterone hapten, an artificial antigen, a polyclonal antibody, and a preparation method and application thereof. Progesterone is used as a raw material; the invention provides a synthesis and purification method for preparing the progesterone hapten. The synthesized and purified progesterone hapten is coupled with a carrier protein to obtain a progesterone complete immunogen for immunizing new Zealand white rabbits, and the polyclonal antibody with high titer and strong specificity is prepared; finally, a biotin/streptavidin amplified signal based enzyme linked immunosorbent assay (BA-ELISA) method is established for detecting progesterone. The BA-ELISA immunoassay method established by the invention has the advantages that the IC50 for detecting progesteroneis 1.29ng/mL, the detection linear range is 0.22 to 7.50ng/mL, the detection limit is 0.080ng/mL, and the application prospect is promising.
