Amidines have been investigated for protection of deoxyadenosine, deoxycytidine, 5-methyldeoxycytidine, cytidine, and deoxyguanosine. These amidine-protected nucleosides and their 5'-bis(p-anisylphenyl)methyl derivatives were prepared in high yield (79-100
N6(N-METHYL-2-PYRROLIDINE AMIDINE)DEOXYADENOSINE - A NEW DEOXYNUCLEOSINE PROTECTING GROUP
McBride, L. J.,Caruthers, M. N.
, p. 2953 - 2956 (2007/10/02)
The synthesis and characterization of N6(N-methyl-2-pyrrolidine amidine) 2'-deoxyadenosine is described. This amidine is significantly more stable toward acid catalyzed depurination than N-benzoyldeoxyadenosine, is easy to prepare and can be removed rapidly and quantitatively.
