The MAO inhibitor l-deprenyl [14611-52-0] prevented the parkinsonism induced by N-methyl-4-phenyl-1,2,5,6-tetrahydropyridine [28289-54-5], and, in human subjects with parkinsonism, potentiated the effect of levodopa [59-92-7] and increased the longevity. The effect of MAO inhibitors on dopaminergic neuron biochem. in the brain is also described. Thus, MAO inhibitors may retard the degeneration of dopaminergic neurons and thus be beneficial in parkinsonism. .

Behavioral performance was tested in a one-way and in a two-way avoidance system. The two monoamine oxidase inhibitors were given acutely (1 and 10 mg/kg, s.c.) and chronically (1 mg/kg s.c./day, for 7 days), and their influence on learning and retention performance was tested 24 h after the last injection. Behavioral reactions remained unchanged after acute administration of the inhibitors, while they were improved following the chronic treatments, either in the one-way or in the two-way experimental situation. Biochem. anal. indicated dissocn. between the influence on striatal dopamine [51-61-6] content and the behavioral effects of deprenyl and clorgyline.
