Chemother., Proc. Int. Congr. Chemother., 14th, Volume Anticancer Sect. 1, 375-6. Edited by: Ishigami, Joji. Univ. Tokyo Press: Tokyo, Japan. (English) 1985. CODEN: 55GNAX. DOCUMENT TYPE: Conference CA Section: 1 (Pharmacology) A method has been developed which allows a rapid spectrophotometric assessment of the in vitro effects of antineoplastic agents on human tumor cells via in situ redn. of 3-(4,5-dimethylthiazol-2-yl)diphenyltetrazolium bromide (MTT) [298-93-1]. The no. of viable cells and the percentage of viability is accurately reflected by the extent of MTT-formazan [23305-68-2] prodn., as measured by an increase in the absorbance at 540 nm. The assay procedure is subjected to a limited no. of causes of failure and has a success rate of 60% per sample received and 80% per test started. The test results show a strong a posteriori correlation between in vitro resistance and clin. resistance. With respect to sensitivity, no false predictions have been obsd. thus far.

F.; Drasar, B. S. (Dep. Med. Microbiol., London Sch. Hyg. Trop. Med., London WC1E 7HT, UK). Food Chem. Toxicol., 24(6-7), 589-90 (English) 1986. CODEN: FCTOD7. ISSN: 0278-6915. DOCUMENT TYPE: Journal CA Section: 4 (Toxicology) Section cross-reference(s): 9 Cell lines (Vero, CHO, chimpanzee liver, HeLa S3, MR5, and Int 407) established in subconfluent monolayers in microtiter plates were exposed 18-20 h to Clostridium perfringes enterotoxin, Escherichia coli Vero toxin, cholera toxin, dibutyryl cAMP [362-74-3] or theophylline (I) [58-55-9], and the plates were processed through a series of stainings for dehydrogenase [9035-82-9], proteins, RNA, and DNA (using methylthiazolyl di-Ph tetrazolium (MTT) [298-93-1], Naphthol Yellow S [846-70-8], pyronin Y [92-32-0], and ethyl green [14855-76-6], resp.). The toxins and chem. affected staining properties of the cells in dose-dependent fashion. The responses obtained for each activity could be considered as a fingerprint, identifying the nature and quantity of that activity.
