DOCUMENT TYPE: Journal CA Section: 1 (Pharmacodynamics) In the isolated perfused liver, 2 mg of 22b-angeloyloxyoleanolic acid (I) [13224-63-0] dissolved in EtOH produced cholestasis within 45 min while 10 mg of asiatic acid [464-92-6] dissolved in EtOH had little effect on the bile flow rate. Changes in the rate of bile salt excretion closely followed those of the bile flow rate. The cholestasis produced was characterized by alterations of the canalicular membrane, whereas cytoplasmic organisation remained unaffected. In the presence of asiatic acid the canalicular region appeared relatively normal and only minor changes to the cytoplasmic organelles were obsd. I had little effect in the changes in lactate and pyruvate levels caused by the metab. of EtOH. Asiatic acid depressed the pyruvate level which led to an elevated lactate to pyruvate ratio but this had returned to control levels after 2 h. Apparently, the cholestatic triterpene acids exert their effect at the canalicular membrane, but the exact mechanism remains unclear.

In rats, plasma I and II levels increased with time for the first 24 h. While the I and II levels of s.c. tissue and adjacent abdominal muscle were maximal by 1 and 3 h, resp., and decreased slowly thereafter. In mice, 2.1 and 2.2% I and II were absorbed by 3 h from the continuous oil phase and oil-water emulsion, resp. In comparison, the radioactivity after oral administration was greatest in the liver, kidney, and paw muscles.
