After development in CHCl3-MeOH (), the bands contg. the nitrofurans are detected under shortwave UV light, scraped from the plate, and extd. with EtOH. The centrifuged exts. are scanned from 500 to 300 nm on a recording spectrophotometer with a tungsten lamp, and the absorbance max. near 360 nm are used for detn. To compensate for extn. and chromatog. losses and for other interferences, the nitrofuran not present in the sample is added as an internal std. The method is generally not applicable at a level below 0.005%, since detection of the bands becomes difficult.

DOCUMENT TYPE: Journal CA Section: 1 (Pharmacodynamics) The high-performance liq.-chromatog. method described here for the quant. anal. for nitrofurantoin (I) [67-20-9] in urine and plasma involves direct anal. of urine samples and anal. of plasma samples after protein pptn. by methanol. A m Bondapak C18 reversed-phase column was used with methanol:0.01 mol/L Na acetate (), pH 5.0, as the mobile phase. The compd. is monitored at 365 nm on a UV absorption detector. The assay, which requires only 0.2 mL of biol. fluid and shows a linear relationship in the range 0.02 to 200 mg/L, can be performed in 9 min and is reproducible. Results for nitrofurantoin so obtained correlate well with those obtained by the Hyamine 10-X spectrophotometric method, but the present method is more sensitive. With no modification, the present procedure can also be used for nitrofurazone [59-87-0]. The sensitivity, accuracy, and convenience of the method make it suitable for clin. monitoring and pharmacokinetic/bioavailability studies with 5-nitrofuran derivs.
