DOCUMENT TYPE: Journal CA Section: 2 (Mammalian Hormones) Rat calcitonin gene-related peptide (CGRP) [83651-90-5] (1 pmol-1 nmol) injected intracerebroventricularly inhibited gastric acid secretion and the secretory response after intracerebroventricularly administered TRH [24305-27-9] in awake, freely moving, pylorus-ligated rats. CGRP and calcitonin (salmon) inhibited gastric secretion similarly. CGRP given intracerebroventricularly inhibited gastric acid secretion stimulated by pentagastrin [5534-95-2], histamine [51-45-6], or bethane-col [590-63-6] for 2 h in anesthetized rats. Adrenalectomy or noradrenergic blockade with bretylium tosylate did not affect the gastric inhibitory action of CGRP. After subdiaphramatic truncal vagotomy, CGRP did not exhibit the inhibitory effect on gastric acid secretion stimulated by pentagastrin. Thus, CGRP injected intracerebroventricularly is a potent inhibitor of gastric acid secretion in the rat and the inhibition apparently depends on intact vagus nerves not the sympathetic nervous system.

The av. granule diam. was 0.73 mm in fasted animals and was reduced to 0.68 mm after 120 min and 2 i.p. injections of urecholine (I) [590-63-6]. Redn. of granule size was attributable to the preferential discharge of large-size granules. The range between 1st and 3rd quartiles of the distribution curve was identical in the 2 groups at 0.24 mm. There was no significant increase in the proportion of granules <0.40 mm after I stimulation. A 3rd group of animals was hyperstimulated by an infusion of a mixt. of caerulein [17650-98-5], secretin [1393-25-5], and I. After 210 min, the av. granule diam. was reduced to 0.43 mm. The range between 1st and 3rd quartiles of the distribution curve was 0.16 mm. In this group, 43.5% of the granules was <0.40 mm. Granule size redn. was accompanied by the appearance of numerous pleiomorphic condensing vacuoles. Thus, secretagogues cause the preferential release of large-size granules and favor the formation of small-size granules.
