L-Valine Me ester-HCl [6306-52-1] (1.68 g, 10 mmol), 1-hydroxybenzotriazole (1.35 g, 10 mmol), and dicyclohexylcarbodiimide (2.06 g, 10 mmol) were added to a suspension of K isocyanoacetate [58948-98-4] (1.23 g, 10 mmol) in MeCN (20 mL) under ice cooling. The reaction mixt. was stirred for 1 h at the same temp. and AcOEt (20 mL) was added. The insol. materials were filtered off and the filtrate was washed with 0.5M aq. citric acid, satd. aq. NaHCO3 and brine, dried over MgSO4, and then concd. to dryness. The crystals obtained were isolated by suction, washed with diisopropyl ether, and recrystd. from AcOEt-hexane to yield I (1.1 g, 55.6%). Of the 6 isonitrile dipeptides synthesized in a similar manner, I was the most active in germination bioassays. I inhibited germination in rice and cucumber by 80-100% at 100 ppm. .

The peptide tends to adopt an alternating right-handed and left-handed helical conformation. The dehydrophenylalanine side chain is planar whereas the valyl side chain adopts a sterically most preferred conformation. The mols., linked by intermol. hydrogen bonds and van der Waals forces, are arranged in helices along the c-axis. The helices are held side-by-side by van der Waals contacts. .
