PLK inhibitors

PLK (Polo-like kinase) inhibitors are a class of compounds designed to target and inhibit the activity of polo-like kinases, a family of serine/threonine protein kinases crucial for regulating cell cycle progression and mitosis. There are several members in the PLK family, with PLK1 being the most extensively studied due to its pivotal role in controlling processes such as centrosome maturation, spindle assembly, and cytokinesis. PLK inhibitors function by binding to the ATP-binding site of PLKs, thereby preventing the phosphorylation of their substrates and disrupting mitotic progression. In cancer therapy, overexpression or dysregulation of PLKs, particularly PLK1, is often observed and associated with uncontrolled cell proliferation and genomic instability.
