Methods (ILꢀ8)
(1) Cell cultures. Immortalized human Keratiꢀ
nocytes (HaCaT cells) were maintained at 37 C in a
humidified 5% CO2 atmosphere in DMEM medium
that contained 10% FBS, and Penicillin/Streptomyꢀ
cin. The cells were plated in the previous day to treatꢀ
ment in the following day.

(2) Measurement of ILꢀ8 (interleukinꢀ8) secretion.
Overnight plated HaCaT cells in 24ꢀwell plates were
washed two times with PBS and exposed to UVB radiꢀ
ation (30 mJ/cm2). Then the medium was exchanged
for Phenol redꢀfree DMEM that contained 2% FBS,
and Penicillin/Streptomycin in the absence or presꢀ
ence of test substances for 24 h. To determine the levels
of ILꢀ8 secreted into cultured media, the supernatants
were collected and analyzed for the presence of ILꢀ8
using commercial available ELISA kits according to
the instructions from the manufacturer (R&D Sysꢀ
tems). At the same time, cell proliferation assay was
performed with WSTꢀ1 reagent (Roche) and used to
compensate for the substance toxicity.
