5.4. General procedure for the condensation of nitrile and 2-
mercaptoacetic acid
To a solution of nitrile (33a, b, 36aef, i) (1.0 mmol) in pyridine
(2.5 mL) was added 2-mercaptoacetic acid or 37aec (1.0 mmol),
and the mixture was stirred at reflux for 8 h. After addition of H2O
(10 mL), the mixture was extracted with EtOAc (3 ꢁ 10 mL) and the
combined organic layers were washed with brine (30 mL) and dried
(MgSO4). Evaporation of the solvent followed by chromatography
(hexaneeEtOAc, 1:1) of the residue gave thiazole (5, 6, 10, 14, 16e20
and 24).
