Described herein are the syntheses of a variety of
templates derived from pyrrolidine trans-lactams. We
found much of the chemistry of the trans-lactam ring
system to be governed by the planarity of the bicylic
template, the propensity for lactam opening or equili-
bration to the corresponding cis-fused system. These
templates have a range of inhibitory activities against
HNE. The details of activity and further medicinal
chemistry of these templates will be described
elsewhere.
