(90:10!85:15)] aorded impure sweroside (2a) (140 mg)
and a mixture of other compounds unidenti®ed as yet
(280 mg). Further separation on the chromatotron
[CH2Cl2±MeOH (93:7!90:10)] gave pure sweroside
(2a) (77 mg).

3.4. Studies on loganin
3.6. Sweroside aglucone-1-acetate (3)
The properties of loganin (above, mp 220ꢀC) were
identical in all aspects (NMR, mass spectra) to values
published in the literature (Boros and Stermitz, 1990).
The optical rotation, [a]2D5=À62.4ꢀ (c=0.0004, MeOH),
is somewhat lower than the literature value of À82.8ꢀC
(El-Naggar and Beal, 1980).
Loganin (1a) (230 mg) was suspended in water pH 6.8
(5 ml) and b-glucosidase (30 mg) added. The solution
was stirred at 37ꢀC for 5 h, then exhaustively extracted
with ether to give impure loganin aglucone. The latter
was puri®ed on the chromatotron [(CH2Cl2±Et2O (5:4)]
to give pure loganin aglucone (1b) (98 mg), oil. 1H
NMR (200 MHz, CDCl3): ꢀ=1.15 (3H, d, J=6.6 Hz,
Me), 1.53 (1H, m, H-6a), 2.30 (1H, m, H-6b), 1.95 (1H,
m, H-8), 1.95 (1H, m, H-9), 3.17 (1H, m, H-5), 3.72 (3H,
s, CO2Me), 4.13 (1H, m, H-7), 4.98 (1H, d, J=4.9 Hz,
H-1), 7.41 (1H, d, J=1.3 Hz, H-3). 13C NMR (50 MHz,
CDCl3): ꢀ=13.3 (C-10), 31.5 (C-5), 41.4 (C-6), 42.0 (C-
8), 46.2 (C-9), 51.4 (OMe), 74.5 (C-7), 95.6 (C-1), 115.5
(C-4), 151.7 (C-3), 168.2 (C-11).
Loganin aglycone-1-monoacetate 1(c). Loganin agly-
cone (43 mg) was treated with DABCO (6 mg) and
shaken to dissolve in THF (2.5 ml). Ac2O (0.2 ml) was
added and the solution stirred at RT overnight.
Removal of solvent, followed by separation on a small
column (silica gel) eluted with CHCl3, followed by
CH2Cl2±Et2O (7:3) gave a pure oil, (35.4 mg). EI±MS
m/z (rel. int.): 270 (M+, 2.1), 210 (M-OAc, 19), 192 (M±
OAc±H2O, 22), 150 (21), 139 (100), 125 (24), 97 (19). ꢀ H
NMR (200 MHz, CDCl3): ꢀ 1:15 (3H, d, J=6.8 Hz,
Me), 1.80 (1H, m, H-6a), 2.11 (3H, s, OCOMe), 2.20
(2H, m, H-8, H-9), 2.32 (1H, m, H-6b), 3.18 (1H, m, H-
5), 3.73 (3H, s, CO2Me), 4.14 (1H, m, H-7), 6.07 (1H, d,
J=3.5 Hz, H-1), 7.32 (1H, d, J=0.8 Hz, H-3).
Sweroside aglucone (2b) (above, 45 mg) was treated
with DABCO (6.2 mg), THF (2.5 ml) and acetic anhy-
dride as detailed for loganin aglycone to give the 1-
acetory derivative (3) (31 mg), mp 112±113ꢀC, (obtained
only after several recrystallisations from 95% EtOH),
[a]2D0 36:4ꢀ (c=0.112, CHCl3); IR (KBr): ꢂ : 3160,
ꢁmu
1765, 1701, 1618, 1404, 1284, 1201, 1189, 1060. EI±MS
m/z (rel int.): 238 (M+, 2.4), 196 (1.4). 178 (2.6), 160

(8.5), 127 (100), 112 (2.5), 97 (8.8). H and 13C NMR
(500 and 125 MHz, CDCl3) are collected in Table 1.
For the X-ray study on (3) diraction data were col-
lected on a Nonius CAD 4 diractometer with graphite
monochromated MoK radiation. The unit cell para-
meters were determined by a least squares ®t. The
structure was resolved by direct methods. Further
details for 1-acetoxy sweroside aglucone (3) are:
C12H14O5. Final R value 0.0470, colourless orthorhom-
bic crystals, cell parameters (Aꢀ) a=7.9637 (7),
b=10.2678 (8), c=29.326 (2), crystal dimensions (mm)
0.58Â0.46Â0.35, space group P212121, V(Aꢀ ) 2398.0(3),

Z=8. (Fig. 1).
The dihedral angles H-1/C-1±C-9/H-9 and H-5/C-5±
C-9/H-9 were 177.9 and 174.5ꢀ respectively, in good
accord with the stereochemistry predicted by the 1H
NMR data.
