Some new 7,9-disubstituted 7H-1,2,3,4-tetrazolo[1,5-c]pyrrolo[3,2- e]pyrimidines 5 have been synthesized either by diazotization of 4-hydrazino- 5,7-disubstituted-7H-pyrrolo[2,3-d]pyrimidines 4 obtained by hydrazinolysis of 4-chloro-5,7-disubstituted-7H-pyrrolo[2,3-d]pyrimidines 3 or via a substitution reaction between 3 and sodium azide. 5,7-Disubstituted-7H- pyrrolo[2,3-d]pyrimidin-4(3H)-ones 2 were obtained by cyclocondensation of 2- amino-3-cyano-1,4-disubstituted pyrroles 1 with formic acid which on chlorination using phosphorus oxychloride afforded 3. A novel route for the synthesis of 4-amino-5,7-disubstituted-7H-pyrrolo[2,3-d]pyrimidines 6 by the reductive ring cleavage of 5 has been reported.

3-Amino-5-aryl-4-imino-7-(p-tolyl)-4,7-dihydropyrrolo-3H-pyrimidines 3 were synthesized from reacting 2-ethoxymethylideneaminopyrrole-3-carbonitriles 2 with hydrazine hydrate.Rearrangement of 3 to 4-hydrazino-7H-pyrrolopyrimidines 4 was carried out.Products 3 were cyclized to the corresponding substituted 9-aryl-7-(p-tolyl)-7H-pyrrolotriazolopyrimidine derivatives 6-12 and 14 upon reacting with triethyl orthoformate, acetic acid, aldehydes, ethyl chloroformate, acyl chlorides and isothiocyanates respectively.The N-substituted-N'-pyrimidin-3-yl>ureas 13a-c and thioureas 13d-h were also obtained from reacting 3a with isocyanates and isothiocyanates respectively.Condensation of 4b with aldehydes and ketones afforded the hydrazones 15 in good yields. - Key words: Pyrrolopyrimidines; pyrrolotriazolo-pyrimidines.
PYRROLOPYRIMIDIMES. I: Synthesis of Novel Pyrrolopyrimidine Derivatives With Antimicrobial Activity

The preparation and cyclization of 2-aminopyrrole-3-carbonitriles 3 to the corresponding pyrrolopyrimidin-4-ones 5 were carriedout.The products 5 with phosphoryl chloride gave 4-chloropyrrolopyrimidine derivatives 7, which when reacted with hydrazine hydrate gave the corresponding 4-hydrazinopyrrolopyrimidines 8 in excellent yield.The syntheses of triazolo-9, tetrazolo-10 and 4-(3,5-dimethylpyrazolo)-12 pyrrolopyrimidines, as well as of a 3-(pyrrolopyrimidynylhydrazono)butanoate ester 13, are also reported.Selected examples from the newly synthesized compounds were tested in vitro for their antimicrobial activity.
