A new family of non-nucleosidic phosphoramidite reagents derived from either esters or amides of 2,2-bis(hydroxymethyl)malonic acid is described.The utility of the building blocks 1-5 is demonstrated by preparation of 5'-phosphorylated oligonucleotides as well as hydrophobic, polyamino and fluorescent labelled oligonucleotide analogues.
