Description
Propiverine hydrochloride is a new urological agent developed for pollakuria and
aconuresis. It appears to exhibit anticholinergic properties and in animal experiments
inhibited KCl induced contractions in the bladder and ileum.

General Description
Propiverine hydrochloride drug is a tertiary amine with spasmolytic and analgesic properties. This drug exerts neurotropic and musculotropic effects on the urinary bladder and smooth muscles. Propiverine hydrochloride has a half-life of 20 hours.
Biochem/physiol Actions
Propiverine is an antimuscarinic drug used for the treatment of overactive blaqdder and urinary incontinence. Propiverine and/or its metabolites also have calcium channel blocking properties in addition to antimuscarinic activity, which may contribute to the clinical activity.

Metabolism
Propiverine is extensively metabolised by intestinal and
hepatic enzymes. Four metabolites were identified in
urine; two of them are pharmacologically active and may
contribute to the therapeutic efficacy.
Propiverine and its metabolites are excreted in the urine,
bile and faeces.

The invention relates to a preparation method of propiverine hydrochloride; a compound represented by the formula I and 1-methyl-4-piperidinol are subjected to transesterification, and then are subjected to hydrochlorination to generate the product propiverine hydrochloride. The used raw materials and the reaction route are all simple and easy to obtain. More importantly, through scale-up experiment of the route, the yield and purity of the product obtained by the preparation method are found to have little difference from those of a product obtained in laboratories, and the preparation methodis suitable for industrialized popularization.
