Synthesis, base pairing properties and nuclease resistance of oligothymidylate analogs containing methoxyphosphoramidate internucleoside linkages

A dithymidine N-methoxyphosphoramidate was synthesized and incorporated into oligothymidylate analogs. The analogs form less stable duplexes with poly dA and poly rA than the corresponding 'all diester' oligomer. The instability is more pronounced with poly rA than with poly dA. The analogs were found to be highly resistant to nucleases.
