APPLICATION: DE 75-2533048 24 Jul 1975. DOCUMENT TYPE: Patent CA Section: 62 (Essential Oils and Cosmetics) Section cross-reference(s): 28 Spiroketals of cyclic ketones were prepd. and used as inflammation inhibitors in compns. for prevention and treatment of sunburn. For example, 16.8 g cyclopentanone [120-92-3] was condensed with 29.2 g 2,2,4-trimethylpentan-1,3-diol [144-19-4] and 29.6 g CH(OEt)3 to give 27.3 g 7-isopropyl-8,8-dimethyl-6,10-dioxaspiro[4.5]decane (I) [62406-73-9]. An oral dose of 1000 mg I/kg decreased carrageenin- and dextran-induced foot edema in rats by 66 and 27%, resp., and a 5% petrolatum soln. or tincture of I decreased UV-induced edema and erythema in mice and guinea pigs, resp., by 44 and 33% after 30 h. I had oral LD50 of >1.0 g/kg. An analogous compd., 2,8,8-trimethyl-7-isopropyl-6,10-dioxaspiro[4.5]decane [62406-75-1] (20 g) and 30 g Me salicylate were suspended in 100 g paraffin oil and then mixed with 300 g lecithin-contg. vegetable oil, 400 g olive oil, 100 g iso-Pr myristate, and 100 g purcellin oil to give a sun-protective oil. .

- Thienyl-substituted cyclic acetals and ketals as sunburn protectants for cosmetic preparations. Moeller, Hinrich; Gloxhuber, Christian (Henkel und Cie. G.m.b.H., Ger.). Ger. Offen. DE 2523129 9 Dec 1976, 13 pp.Some commonly used reagents like 98-03-3 and 62435-24-9 are used in this experiment. (German). (Germany). CODEN: GWXXBX. CLASS: IC: A61K007-00. APPLICATION: DE 75-2523129 24 May 1975. DOCUMENT TYPE: Patent CA Section: 62 (Essential Oils and Cosmetics) Section cross-reference(s): 28 The title compds. are inflammation inhibitors esp. for use in sunscreens and for treatment of sunburn. Thus, UV-induced erythema in guinea pigs was inhibited by 45% by painting the affected area 8 times over 6 h with 5% tincture of 2-(2-thienyl)-4-isopropyl-5,5-dimethyl-1,3-dioxane (I) [62435-24-9]. I was prepd. by heating a mixt. of 40.0 g thiophene-2-aldehyde [98-03-3], 53.0 g 2,2,4-trimethylpentane-1,3-diol [144-19-4], and 0.3 g p-toluenesulfonic acid in 250 mL C6H6 under reflux; the product was purified by washing with aq. NaHCO3 and water, removal of C6H6, and fractional distn. A sunscreen oil was prepd. by suspending 2-(5-methyl-2-thienyl)-4-isopropyl-5,5-dimethyl-1,3-dioxane [62435-25-0] 20 and menthyl salicylate 30 in paraffin oil 100 g with heating and adding lecithin-contg. plant oil 300, olive oil 400, iso-Pr myristate 100, and purcellin oil 100 g at room temp. .
