DOCUMENT TYPE: Journal CA Section: 1 (Pharmacodynamics) I.v. administration of RGH-4405 (I) [42971-09-5], and RGH-4406 (II) [19877-89-5] to rats at 1 mg/kg dose significantly improved the ischemic anoxia of brain as measured by the prolongation of the duration of cortical resistance time (time between occlusion of arteries and the onset of silence brain elec. activity). Vincamine (III) [1617-90-9] showed similar effects at 2 mg/kg dose level, but not at 1 mg/kg. All 3 alkaloids decreased the recovery time, i.e., the time for reappearance of electroencepholog. activity on removal of the arterial occlusion. The increase tolerance to anoxia after administration of these drugs may be due to an increase in brain O utilization.

However, when administered as i.v. infusions (10 mg/kg/min) all 3 drugs increased the brain microcirculation. Tachyphlaxis was obsd. after repeated administration of the drugs.
