DOCUMENT TYPE: Journal CA Section: 1 (Pharmacology) A Widal-type agglutination bioassay modified for measurement of roxithromycin (I) [80214-83-1] in human plasma, urine, milk and mouse plasma was developed. Bioassay plates were prepd. using Antibiotic Medium No. 11 (pH 8.0) and Sarcina lutea as test organism. Both urine and plasma assays consistently detected I concns. 31.0 mg/mL while in some human milk samples and mouse plasma concns. as low as 0.25 mg/mL were detected. The accuracy and precision of the I assay procedures are satisfactory for most clin. applications.

DOCUMENT TYPE: Journal CA Section: 64 (Pharmaceutical Analysis) Roxithromycin and its related substance in tablets were detd. by HPLC on ODS column (250 mm x 4.6 mm. 5 mm) with 0.067 mol L-1 ammonium dihydrogen phosphate soln. (adjusted pH to 6.5 by triethylamine) and acetonitrile (3:2) as mobile phase. The flow rate was 1 mL min-1 and the detective wavelength was 210 nm. The content of roxithromycin and its related substance were calcd. with the external std. method. The linear range of roxithromycin was 0.5 - 4.0 mg mL-1 with r = 1.0. The linear range of the related substance was 0.5 - 50 mg mL-1; the limit detection was about 0.01 mg and the LOQ was about 0.04 mg. The method was accurate, simple and quick and could be used to analyze roxithromycin. .
