Metabolites produced during a 4 h incubation period were isolated by HPLC and identified by NMR-, IR-, UV-spectrometry, and mass spectroscopy. SPIR was not metabolized in a detectable amt. CAN was converted to canrenoic acid [4138-96-9], several hydroxylated (15b- [65125-84-0], 16a- [92219-03-9], 19- [66878-34-0], 20R- [92343-27-6], and 21S-hydroxy-CAN [92282-12-7]), and 1 reduced metabolite (3a-hydroxy-CAN [92283-64-2]). When DHC was incubated, an analogous pattern was detected. Thus, CAN and DHC serve as substrates for steroid metab. in the rat testis.

In human heart muscles, Aldactone and canrenone also exerted a concn.-dependent neg. inotropic effect; at 1,000 mmol/L and 250 mmol/L, they reduced force of contraction by 57 and 67%, resp.
