JAK inhibitors

Janus kinase (JAK) inhibitors are a class of pharmaceutical compounds that target and inhibit the activity of Janus kinases, a family of intracellular enzymes crucial for signaling pathways involved in immune responses and hematopoiesis. JAKs are associated with cytokine receptors on the cell surface, including those for interleukins and interferons. When cytokines bind to their receptors, JAKs become activated and phosphorylate signal transducers and activators of transcription (STATs), which then translocate to the nucleus and regulate gene expression. By inhibiting JAK activity, JAK inhibitors interfere with this signaling cascade, modulating immune responses and inflammation.
