 Synthesis of Ketanserin
  • Synthesis of Ketanserin
  • (CAS NO.: ), with its systematic name of 2,4(1H,3H)-dione, 3-(2-(4-(4-fluorobenzoyl)-1-piperidinyl)ethyl)-, could be produced through many synthetic methods.

    Following is one of the synthesis routes: 
    N-(2-bromoethyl)-2-nitrobenzamide (I) is condensed with 4-(4-fluorobenzoyl)piperidine (II) in the presence of Na2CO3 in refluxing methyl isobutyl ketone to produce N-[2-[4-(4-fluorobenzoyl)-2-piperidinyl]ethyl]-2-nitrobenzamide (III), and the yielding compound is reduced with H2 over Pt/C in methanol yielding the corresponding amino derivative (IV). Finally, this compound is cyclized with urea (V) in refluxing xylene.
