  • (CAS NO.: ), with its systematic name of N-(1-Methyl-2-piperidinoethyl)-N-(2-pyridyl)propionamide fumarate, could be produced through many synthetic methods.

    Following is one of the synthesis routes:

    Synthesis of Propiram fumarate

    1-(1-Piperdinyl)acetone oxime (I) is reduced LiAlH4 to produce 1-methyl2-(1-piperidinyl)ethylaine (II) in the presence of ethyl ether, THF or dioxan, and then the compound is acylated with propionyl chloride (III) in ether to afford propionamide (IV). Next, compound (IV) is condensed with 2-bromopyridine (V) by means of K2CO3 and Cu (dust) to afford compound (VI) with the condition of heating. Alternatively, (VI) can be obtained by heating N-[1-methyl-2-(1-piperidinyl)ethyl]-N-(2-pyridyl)amine (VII) with propionic anhydride (VIII). Finally, heating derivative (VI) with fumaric acid in acetone/EtOH allows formation of the desired compound as the corresponding fumarate salt.
