Complete automatic sequence analysis of peptides may be achieved using aromatic isothiocyanate derivatives containing sulfonic acid groups to label the ε amino groups of lysine residues. Two new fluorescent derivatives V and VI (isothiocyanates of 2 phenylbenzothiazol and pyrene) are described. Using ε labeling and the protein program of the sequenator, it was possible to analyze automatically the sequences of the insulin peptide chains.
