A simple and environmentally friendly strategy for the synthesis of E-ortho-hydroxystilbenes has been established. Two kinds of α,β-unsaturated aromatic lactones, i.e. the 3-arylcoumarins and the isoaurones, could both readily undergo a cascade hydrolyzation/decarboxylation reaction in the presence of KOH in ethylene glycol to afford the desired E-ortho-hydroxystilbenes in moderate to high yields.

Since the past decades, the plant stilbenoid resveratrol has gained significant attention of the general public as well as the research community due to its versatile medicinal properties. Apart from resveratrol, there is also an increasing interest in other plant stilbenoids because of their different potential biological activities. In order to meet the increasing demand for stilbenoids, alternative and sustainable approaches for their production are needed. We identified the cytochrome P450 monooxygenase 154E1 from Thermobifida fusca YX (CYP154E1) which enables the synthesis of (E)-4,4′-dihydroxystilbene via direct double hydroxylation of (E)-stilbene. The construction of a triple mutant led to a more than six-fold increased catalytic efficiency compared to the wild type enzyme. CYP154E1 and variants thereof accepted not only (E)-stilbene but also possessed remarkable activity towards ortho- and meta-substituted hydroxystilbenes leading to resveratrol, (E)-2,4′-dihydroxystilbene, (E)-2,4′,5-trihydroxystilbene and (E)-3,4′-dihydroxystilbene. The combination of protein engineering and the use of methyl-β-cyclodextrin as substrate solubilizing agent resulted in product titers of up to 4.2 g L?1 and enzyme total turnover numbers (TTN) of up to 20,000. A facile and rapid access to resveratrol derivatives has been achieved based on palladium-catalyzed oxidative Heck reaction of aryl boronic acids with styrenes followed by demethylation in moderate to good yields. A series of resveratrol derivatives with various functional groups has been synthesized easily. The radioprotective activity of synthesized compounds has also been evaluated using rat thymocytes. The results revealed that some resveratrol derivatives efficiently protected the thymocytes from radiation-induced apoptosis.
Trans-O-hydroxy-diphenyl ethylene derivatives and its preparation method and application

The invention discloses a trans-o-hydroxy stilbene derivative as well as a preparation method and application thereof. The preparation method comprises the following steps: in the presence of a rhodium catalyst and acetate, reacting an o-hydroxy styrene compound with arylboronic acid in a solvent, and after the reaction is completed, treating, thereby obtaining the trans-o-hydroxy stilbene derivative. The preparation method is mild in reaction conditions, procedures are simple and can be rapidly performed, and the yield is relatively high. The cell experiment shows that compared with resveratrol, a product prepared from the trans-o-hydroxy stilbene derivative is generally better in anti-tumor activity.

A simple synthesis of 2-hydroxylated (E)-stilbenes was accomplished in good yields via oxidative coupling of 2-hydroxystyrenes and arylboronic acids, with Rh(III)-catalyst and Cu(OAc)2 as oxidant. The antiproliferative evaluation of all the synthesized compounds were assessed on four different human cancer cell lines (Colo-205, MDA-468, HT29, and MGC80-3), and the results showed that several compounds exhibit strong antiproliferative activities (up to IC50 = 35 nM for MGC80-3).

Various (E)-hydroxystilbenes were synthesized from (E)/(Z) mixtures of methoxystilbenes through a new (Z)-(E) isomerization method followed by demethylation. The nematocidal activity appears when methoxystilbenes are demethylated to hydroxystilbenes. For this activity, a hydroxy group at the C-2 or C-3 position is necessary. Thus, 2-hydroxy-, 3-hydroxy-, 2,6-dihydroxy-, 3,4-dihydroxy-, 3,5-dihydroxy-, 2,2'-dihydroxy-, 3,3'-dihydroxy-, 3,4'dihydroxy-, 2-hydroxy-4-methoxy-, 5-hydroxy-2-methoxy-, 2-hydroxy-6-methoxy-, 6-allyloxy-2-hydroxy-, 3-hydroxy-5-methoxy-, and 5-allyloxy-3-hydroxystilbenes showed rather potent nematocidal activity. The activity of 5-allyloxy-3-hydroxystilbene was the strongest [minimal lethal concentration (MLC) = 30 μM]. The activities of the (E) and (Z) isomers were comparable. The activities were also retained, though they were weaker, in the dihydro derivatives, hydroxybibenzyls.

Sixteen (E)-2-hydroxy-4'-substituted stilbenes have been prepared by the Wittig reaction.Direct coupling of the appropriate p-substituted benzylidenetriphenylphosphoranes with salicylaldehyde afforded the (E)-stilbenols (2) 4'-R = H(2a), Me(2b), But(2c), Ph(2d), CN(2e), NO2(2i), OMe(2k), F(2n), Cl(2o), and Br(2p).Alternatively, the Wittig reaction of o-methoxymethoxybenzylidenetriphenylphosphorane with various p-substituted benzaldehydes gave the (E)-methoxymethylstilbenols (4) which upon acidic cleavage furnished the corresponding stilbenols (2) 4'-R = CN(2e), CO2Me(2f), NMe2(2h), OH(2j), OAc(2m), and I(2q).The symmetrical (E)-2,2'-dihydroxystilbene was obtained in an analogous fashion.Reduction of nitrostilbenol (2i) yielded the corresponding aminostilbenol (2g).
