Synthesis, characterization and studies on the biological activity of bis-cyclometalated M(III)-complexes (M = Rh, Ir and Ru)

The synthesis and characterization of new cyclometalated complex salts [M(ptpy)2(4,4́-HOCH2bpy)]PF6 (M = Rh, 1; M = Ir, 2 and M = Ru, 4; ptpy = 2-(p-tolyl)pyridinato; 4,4́-HOCH2bpy = 4,4́-bis(hydroxymethyl)-2,2́-bipyridine) is reported. Compounds 1 and 2 were obtained by bridge-splitting reaction of complexes [{M(µ-Cl)(ptpy)2}2] (M = Rh, Ir) with the ligand 4,4́-HOCH2bpy in refluxing CH2Cl2/MeOH/H2O mixtures. Compound 4 was obtained by a multi-step synthesis starting with the precursor compound [{Ru(µ-Cl)Cl(η6-C6H6)}2]. All compounds were comprehensively characterized (elemental analysis, MS, IR, UV–Vis, and 1H and 13C NMR where appropriate), and the molecular structures of compounds 1 and 4 were confirmed by X-ray diffraction. All new compounds display significant cytotoxicity against human cancer cell lines with the IC50 values in the low micromolar range. Quite surprisingly, there seems to be no significant influence of the central metal on the biological activity.
