Phaseollin (cas 13401-40-6) metabolism and tolerance in Fusarium solani f. sp. phaseoli

Metabolism of the bean phytoalexin, Phaseollin (cas 13401-40-6), to 1a-hydroxyphaseollone by Fusarium solani f. sp. phaseoli appears to occur by way of an inducible mono-oxygenase. Evidence for this conclusion is that: (1) pretreatment with selected aromatic compounds, as well as phaseollin itself, enhanced the ability of the fungus to metabolize phaseollin, (2) the enhanced ability to metabolize phaseollin did not occur in cultures previously treated with cyclohexi-mide, an inhibitor of protein synthesis, (3) naphtho-(2,3-d)-1,3-dioxole, an inhibitor of mono-oxygenases, significantly decreased the rate of phaseollin metabolism and (4) oxygen from O2 but not from H2O was incorporated into 1a-hydroxyphaseollone during the metabolism of phaseollin.Phaseollin metabolism may be a component of, but is not totally responsible for, adaptive tolerance to phaseollin. The expression of tolerance to phaseollin was closely correlated in time with the metabolism of phaseollin. In addition, pretreatment of the fungus with compounds that stimulated phaseollin metabolism also enhanced phaseollin tolerance. However, inhibition of phaseollin metabolism by naphtho-(2,3-d)-l,3-dioxole did not render cultures completely sensitive to the toxic effect of the phytoalexin.
