Some pharmacologic properties of benzphetamine hydrochloride

Some comparisons of the pharmacologic properties of benzphetamine hydrochloride, amphetamine sulfate, and phenmetrazine hydrochloride are reported. Acute anorexigenic studies in dogs indicated that benzphetamine 1 mg/kg is equivalent to d-amphetamine 0.2 mg/kg or to phenmetrazine 14 mg/kg. Analysis of data from chronic anorexigenic experiments showed that benzphetamine is approximately one-third as potent as d-amphetamine and 10 times as potent as phenmetrazine.Benzphetamine was found to be about one-eighth as stimulant as d-amphetamine on motor activity in mice. To produce equivalent peak pressor effects in anesthetized dogs with benzphetamine, 20 times the amphetamine dose was needed. The motor activity and blood pressure studies tended to indicate that benzphetamine has a longer duration of action than amphetamine. In toxicity studies it was observed that there is less difference in lethality between grouped versus individual mice with benzphetamine and phenmetrazine than with d-amphetamine.
